Öğe Cephalaria duzceensis (Dipsacaceae), a new species from the western Black Sea region, Turkey(Wiley, 2007) Aksoy, Necrni; Göktürk, R. Süleyman; Açık, Leyla; Çelebi, AytenA new species, Cephalaria duzceensis N. Aksoy & R. S. Gokturk (Dipsacaceae) from western Black Sea region of Turkey is described. It is illustrated in line drawings and the morphology of the new species is compared with Cephalaria speciosa. Molecular and morphological methods were used to separate the two species. Both species were first identified morphologically and then studied by randomly amplified polymorphic DNA (RAPD) markers. Genetic similarities were calculated based on the RAPD data and used to construct a UPGMA dendrogram. According to the result, two main clusters were observed using the character differences. It is concluded that C duzceensis sp. nov. is different from C. speciosa.Öğe Characterization of Staphylococcus aureus strains isolated from subclinic bovine mastitis by protein patterns, antibiotic resistance and plasmid profile(2006) Aslantaş, Özkan; Küp, Fatma Öztürk; Çelebi, Ayten; Açık, Leyla; Ergün, YaşarHatay bölgesindeki subklinik inek mastitislerinden izole edilen toplam 50 Staphylococcus aureus susunun protein paterni, antibiyotik dirençliliği ve plazmid profili yönünden karakterizasyonu yapıldı. S. aureus suşlarının SDS-PAGE ile analizinde moleküler ağırlığı 14.4-116 kDa arasında değişen 17-49 bant elde edildi. Elektroforetik protein bantlarının bilgisayar destekli numerikal analizi sonucunda S. aureus suşları 2 gruba ayrıldı. S. aureus suşları trimethoprim-sülfametoksazol, danofloksasin, amoksisilin-klavulanik asit, sulbaktam-ampisilin, eritromisin, linkomisin, sefoperazon, oksitetrasiklin, amoksisilin, streptomisin ve penisilin'e sırasıyla % 4, % 4, % 10, % 14, % 30, % 34, % 36, % 38, % 50, % 54 ve % 68 oranlarında dirençli bulundu. İncelenen izolatların 47'sinde, büyüklüğü 1.7-24 kb arasında değişen plazmidler saptandı.Öğe Protein patterns and plasmid profiles of the bacterial strains isolated from a poultry slaughterhouse in Ankara, Turkey(Faculty Food Technology Biotechnology, 2005) Açık, Leyla; Temiz, Ayhan; Çelebi, Ayten; Arslan, Sevim; Yılmaz, RemziyeA total of 65 identified isolates of coliform bacteria Salmonella, including Campylobacter and Staphylococcus isolated from different control points of a poultry slaughterhouse in Ankara, Turkey were characterized by morphological, biochemical and physiological tests including API 10 S system, and by plasmid profiles on agarose gel electrophoresis and whole-cell protein patterns on SDS-PAGE. Plasmids were detected in 53.8 % of the isolates. The molecular mass of the plasmids was within the range from 0.66 to 12.66 mDa. Electrophoretic banding patterns showed that whole cell protein profiles differed in several protein bands in Salmonella, Campylobacter and Staphylococcus species, but the differences were insufficient for reliable differentiation of bacteria species by SDS-PAGE method.Öğe Synthesis, Cytotoxicity, and DNA Interactions of New Cisplatin Analogues Containing Substituted Benzimidazole Ligands(Amer Chemical Soc, 2009) Gümüş, Fatma; Eren, Gökçen; Açık, Leyla; Çelebi, Ayten; Öztürk, Fatma; Yılmaz, Şükran; Elerman, YalcinSix new platinum(II) complexes with 1-H or methyl-2-chloromethyl or acetoxymethyl or 2'-hydroxyethylbenzimidazole carrier ligands were synthesized and evaluated for their reactivity against model nucleophile I-, cellular uptake, and in vitro anti proliferative activities against the human MCF-7 breast and HeLa cervix cancer cell lines. The effect of the compounds on pBR322 plasmid DNA was studied by gel electrophoretic mobility measurements. Flow cytometric analysis was also carried out to study the effect of representative compounds 1 and 2, bearing 2-chloromethyl or -acetoxymethylbenzimidazole carrier ligands, on the cell cycle distribution of MCF-7 and HeLa cells, respectively. In general, it was found that Pt(II) complexes were less cytotoxic than cisplatin and were comparable to carboplatin. The results of the plasmid DNA interaction and the restriction studies suggest that changing the chemical structure of the benzimidazole ligands may modulate DNA binding mode and the sequence selectivity. Compounds I and 2 had no significant effect on the cell cycle profile of the cells used. However, Compound 2 induced a significant increase in the SubG1 cell population at a concentration of 20 mu M.
