Apoptosis-necrosis, genotoxicity, hemolysis, and cell attachment studies are carried out within the scope of in vitro tests, and irritation, sensitivity, and burn wound studies are carried out within the scope of in vivo tests. The apoptotic index value of CA-AgNPs-[PCL/PEO] NFs material on L929 fibroblast cells is determined as 5.0 +/- 1.0% at the highest concentration and the necrotic index value is 5.0 +/- 0.3%. Micronucleus rates (%) of NFs treated with CHO (Chinese Hamster Ovary) cells are not at genotoxic level. The hemolytic index value of NFs dressing is determined as 0.23 +/- 0.03%, The primary irritation index (PII) value of NFs wound dressing is calculated as 0.36 by irritation tests. In addition, the potential sensitization reaction of NFs extract on guinea pigs is evaluated and the sensitization score is determined as 0.9. The healing efficacy of NFs material on second-degree burn wounds compared to a commercial product is supported by pathomorphological findings.Öğe INVESTIGATION OF THE MUTAGENIC, CYTOTOXIC AND ANTIMICROBIAL EFFECT OF BIKAVERIN MYCOTOXIN(Univ Federal Uberlandia, 2024) Beyzi, Ebru; Kulahci, Meryem Burcu; Cerci, Nebahat AytunaBikaverin is a reddish pigment produced by different fungi species (Mycogone jaapii, Verticillium agaricinum, Beauveria bassiana, Paecilomyces fumosoroseus, Polyporus sulphureus), mainly of the Fusarium genus. Due to its pigment feature, bikaverin can be used as a dye in various fields in the industry. However, it is extremely important to study the mutagenic/genotoxic effects, cytotoxic effects and antimicrobial properties of bikaverin for application of industrial areas. In the study, the mutagenic, cytotoxic and antimicrobial effects of bikaverin were investigated. The mutagenic effect of bikaverin was studied with the Ames test. Salmonella typhimurium TA97a, TA98, TA100, TA102 and TA1535 strains were used in the test. Five different doses of bikaverin (0.075, 0.1, 0.2, 0.3 and 0.5 mu g/plate) were tested against strains. It was determined that there was no mutagenic effect of bikaverin. The cytotoxicity of bikaverin was evaluated by MTT test on L929 fibroblast cell line. Bikaverin demonstrated no cytotoxic effect on L929 fibroblast cell line, according to cell viability calculations that showed >73% for all concentrations (1, 0.5, 0.4, 0.3, 0.2, 0.1, 0.075, 0.05, 0.025, 0.01, 0.005 and 0.001 mu g/mL) examined. Bikaverin's IC50 value was determined to be 1.79 +/- 0.51 g/mL. The antimicrobial activity of the bikaverin was evaluated by using the microdilution method. Bikaverin was found to have antimicrobial effects on Methicillin resistant Staphylococcus aureus, Vancomycin resistant Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Candida albicans and Candida krusei, as MIC values ranged from 1.25 -5 mu g/ mL.Öğe Phosphorus-nitrogen compounds: part 68. To prepare these phosphazenes, the starting compounds tetrachloro(4-chlorobenzyl)spiro(N/N)cyclotriphosphazenes, (ClBzSpiro-5)R-1(N3P3)Cl-4 [Bz: Benzyl; R-1: Me (1) and R-1: Et (2)], were prepared regioselectively from the reactions of hexachlorocyclotriphosphazene, N3P3Cl6 (HCCP, trimer) with N-methyl/ethyl-N'-(4-chlorobenzyl)-1,2-diaminoethanes. Reactions of tetrachlorocyclotriphosphazenes (1 and 2) and 9-ethyl-N-methyl-3-carbazolyl-1,2-diaminoethane (3) or 9-ethyl-N-methyl-3-carbazolyl-1,3-diaminopropane (4) produce new cis/trans-dispirocyclotriphosphazenes, [(ClBzSpiro-5)R-1(N3P3)(CzSpiro-n)R-2]Cl-2 (Cz: Carbazolyl; R-1, R-2: Me or Et; n = 5 or 6; (5a-8a and 5b-8b), containing unsymmetrical spiro-architectures. In addition, the structures of trans-7a and cis-7b isomers were clarified by single crystal X-ray crystallography. The chiralities of trans-7a and cis-7b were confirmed using X-ray crystal structures, P-31 NMR spectra recorded upon the addition of chiral solvating agent [(S)-(+)-2,2,2-trifluoro-1-(9 & PRIME;-anthryl)ethanol; CSA], and circular dichroism (CD) spectra. Moreover, the photophysical properties of phosphazenes showed a fluorescence profile with lifetimes of about 4.9-6.6 ns and quantum yields in the range of 0.10-0.14. Additionally, the antibacterial and antifungal activities of the newly synthesized phosphazenes against some bacteria and yeast strains and their interactions with pBR322 plasmid DNA were investigated. The bacterial strain most susceptible (MIC = 156.3 mu M) to compounds 5a and 8a was P. aeruginosa. However, it was found that the yeast strain most susceptible (MIC = 156.3 mu M) to compounds 5b, 7a, 7b and 8b was C.albicans. The cytotoxic activities of 5a, 5b, 7b and 8a against L929 fibroblast and MCF-7 breast cancer cells were determined. However, cis-5b showed reasonable antioxidant activity with a radical scavenging value of 35.20%.Öğe Vitex Agnus-Castus L. Nanoparticles: Preparation, Characterization and Assessment of Antimicrobial and Anticancer Activity(Wiley-V C H Verlag Gmbh, 2023) Ekici, Seda; Bozkaya, Esra; Bozkaya, Ogun; Cerci, Nebahat Aytuna; Aluc, Yasar; Ekici, HusamettinThis study was aimed to synthesise antimicrobial and therapeutic silver nanoparticles (AgNPs) by green synthesis method using Vitex agnus-castus L. (VAC) seed extract. The synthesised VAC seed extract coated AgNPs were characterised by UV-Vis spectrometry, Zeta analysis and FT-IR spectrometry. The antifungal, antimicrobial, cytotoxicity by MTT assay, and anticarcinogenic activities on HeLa cells line were investigated. VACAgNPs exhibited high antimicrobial activity against S. aureus with a zone diameter of 11.3 +/- 0.6 mm. In addition, the cytotoxicity of VAC-AgNPs was evaluated using the L929 fibroblast cell line and the HeLa cells line, with AgNPs applied at six different concentrations. At a concentration of 5.0 mM, viability was found to be 47.6%+/- 4.2% in L929 fibroblast cells and 39.6%+/- 7.9% in HeLa cells, suggesting a potential antiproliferative effect of VAC-AgNPs on HeLa cancer cells. The IC50 values of VAC-AgNPs against the HeLa cell line were calculated as 2.94 mu g/ml.
