Öğe Effect of Epidermal growth factor on ın vitro maturation of cat oocytes recovered from ovaries at follicular and luteal stages(2009) Polat, Bülent; Acar, Duygu Baki; Macun, Hasan Ceyhun; Korkmaz, Ömer; Çolak, Armağan; Baştan, Ayhan; Akçay, AytaçBu çalışmada, seksüel siklusun folliküler ve luteal fazında bulunan kedi ovaryumlarından toplanan oositlerin nükleer maturasyonu üzerinde IVM vasatına çeşitli konsantrasyonlarda katılan EGF’nin etkilerinin belirlenmesi amaçlandı. Çalışmada folliküler ve luteal dönemde bulunan toplam 42 kediden alınan 612 kumulus oosit kompleks kullanıldı. Oositler, (1) TCM 199 (EGF 0); (2) TCM 199 10 ng/ml EGF; (3) TCM 199 50 ng/ml EGF olarak 3 gruba ayrılıp IVM vasatına alındı ve %5 CO2’li ortamda 48 saat süreyle mature edildi. Kromozomal analizlere göre her bir oosit germinal vezikül (GV), Metafaz I-II (M I-II) ve dejenere olarak adlandırıldı. EGF 50 grubunda, luteal dönemdeki ovaryumlardan alınan oositlerin yüksek oranda (%41.7) GV aşamasında kaldığı gözlendi (P0.05). EGF 10 grubunda folliküler dönemdeki oositlerin önemli oranda (%37.9) M II aşamasına ulaştığı saptandı (P0.05). Tüm EGF gruplarında luteal dönemdeki ovaryumlardan alınan oositlerin folliküler döneme göre yüksek oranda M II aşamasına ulaştığı görüldü (P0.05). Sonuç olarak sunulan çalışmada IVM vasatına katılan düşük dozda EGF’nin (10 ng/ml) EGF katılmayan gruba kıyasla maturasyonu olumlu yönde etkilediği gözlendi. Ancak maturasyon vasatına yüksek miktarda (50 ng/ml) katıldığında ise kedi oositlerinin IVM’si üzerinde olumsuz etkilerinin olduğu ve luteal dönemdeki ovaryumlardan toplanan oositlerin folliküler döneme göre IVM için daha uygun olduğu kanısına varıldı.Öğe The Investigation of Serum Estrogen, Progesterone, AMH and IGF-1 Profiles in Lactating and Non Lactating Mares after Altrenogest Administrations(2020) Polat, İbrahim Mert; Korkmaz, Ömer; Elifoğlu, Taha Burak; Pir, İlknur Yağcı; Çolakoğlu, Hatice Esra; Pekcan, MertThe objective of this study is to follow up the follicular growth in ovaries until mating by ultrasonography to determine of pregnancy rates, serum estrogen, progesterone, AMH and IGF-1 concentrations in mares which synchronized with altrenogest and to compare the obtained data with non-lactating mares. The study was carried out during breeding season while ovaries were active; lactating mares were included in Group 1 (n = 8) and non-lactating mares in Group 2 (n = 8). In both groups, synchronization was performed by administration of altrenogest (Regumate®) and prostaglandin F 2 alpha (PGF2?). Dominant follicle diameters of non-lactating mares were found to be greater than those of lactating ones. During the study, estrogen, progesterone, AMH and IGF-1 levels were significantly lower in the lactating mares. The non- lactating mares were found to have a first pregnancy conception rate (75%), similar to that of lactating mares (62%) (x2=1.54, P=0.62). It was concluded that in the mares synchronized with altrenogest, the presence of lactation and the foal are significantly influences ovarian activities, ovarian steroid levels, AMH and IGF-1 levels. Therefore, in order to increase pregnancy rates in lactating mare, sexual cycle synchronization with altrenogest are considered to be a suitable method.Öğe The Effects of Hot Pepper Seeds Added to the Diet in Quails on the Morphology of the Oviduct and Ovary(2023) Korkmaz, Deniz; Kırar, Nurcan; Top, Şermin; Yıldırım, Özge; Aydın, Sadık Serkan; Korkmaz, Ömer; Çetin, MehmetThe aim of this study is to reveal how red-hot pepper seeds added to the diet in quails change ovarian follicle dynamics and the morphological structure of the oviduct. For this reason, a total of 48 female Japanese quails (Coturnix coturnix Japonica) aged 14 weeks were used as a material. While red hot pepper (Capsicum annuum L.) seeds were added to the diet of the animals in the experimental group at the rate of 2%, 4%, and 6%, it was not added to the control group. At the end of the study, while the number of white follicles showed a statistically significant increase in all groups compared to the control group (P<0.05), a significant increase was recorded in the number of yellowish follicles in the groups in which 2% and 4% red pepper seeds were added to the diet compared to the control group. Also, the correlation analysis revealed that there was a positive correlation between the yellowish follicle and the white follicle. In the present study, it was observed that there was a negative correlation between magnum length and isthmus length (r=-0.369, P=0.019), while there was a positive correlation between the total length of the oviduct and yellow follicle diameter (r=0.335, P=0.034). Consequently, it was concluded that the addition of red-hot pepper (2% and 4%) seeds to the diet may increase egg yield in quails.
