Öğe Differences in the Follicular Morphology of Young and Aged Bitches and Their Correlation with the Anti-Mullerian Hormone(Kafkas Univ, Veteriner Fakultesi Dergisi, 2016) Korkmaz, Omer; Korkmaz, Deniz; Polat, Ibrahim Mert; Yagci, Ilknur Pir; Pekcan, Mert; Emre, Birten; Zonturlu, Abuzer KafarThis study aimed to investigate the structural, morphological and cellular differences of follicles at different stages of follicular development as well as to determine the correlation of the ovarian follicle population with serum Anti-Mullerian Hormone (AMH) levels in young and aged bithches. Sixteen bitches were divided into two groups according to their ages. Group A included young bitches aged 2 years (n=8) while Group B constituted of those aged 8 to 10 years (n=8). Diameters of the primordial, primary and preantral follicles were found to be significantly larger in Group B, in comparison to Group A. In Group A, the mean number of granulosa cells was 91.65 +/- 2.23 in the secondary follicles and 301.31 +/- 4.16 in the preantral follicles. In Group B, the same values were found to be 89.46 +/- 2.68 and 270.25 +/- 3.54, respectively. The mean serum AMH levels in Group A and Group B were 0.233 +/- 0.046 ng/mL and 0.099 +/- 0.008 ng/mL, respectively (P<0.05). In conclusion, the results indicated that the number of primordial and primary follicles as well as the numbers of granulosa cells in secondary and preantral follicles decreased with advanced age which resulted in lower serum AMH levels in aged bitches. The results suggested that the AMH, which is used as a fertility parameter in humans, could also be used for the same purpose in dogs.Öğe Effects of injectable trace element and vitamin supplementation during the gestational, peri-parturient, or early lactational periods on neutrophil functions and pregnancy rate in dairy cows(Elsevier, 2021) Yazlik, Murat Onur; Colakoglu, Hatice Esra; Pekcan, Mert; Kaya, Ufuk; Kuplulu, Sukru; Kacar, Cihan; Polat, MertThe aim of this study was to evaluate effects of injectable trace element and vitamin combination on phagocytic, oxidative burst activity of neutrophils and reproductive outcomes in dairy cows. Cows were to assigned to the following groups: (1) injectable trace element supplementation (ITES, n = 44, containing zinc, manganese, copper, selenium); (2) injectable vitamin supplementation (VIT, n = 48, containing vitamins A, D-3, E); (3) ITES + VIT = 46); and (4) control (CON, n = 44). Cows were administered four injections between 230 and 260 days of the gestational period, on day of parturition, and 30 days postpartum. Neutrophil function was assessed at 10 days before and after calving. Phagocytosis was greater in cows of the ITES + VIT group at 10 days prepartum (P < 0.05) while oxidative burst was similar among groups. There were greater non-esterified fatty acid (NEFA) concentrations in cows of the ITES+VIT group at 10 days prepartum (P < 0.05). Cows supplemented with ITES+ VIT had less SOD activity than those supplemented with ITES or vitamin during the pre- to post-partum transition period (P < 0.05). The total odds of pregnancy were greater in cows supplemented with trace element and/or vitamin (P < 0.05). In conclusion, supplementation of ITES and/or VIT resulted in an increased total pregnancy rate. Vitamin or trace element supplementation did not differ with the control group in both the prepartum and postpartum period for immune variables. There, however, was greater phagocytosis in cows supplemented with vitamin and trace elements during the prepartum period that might be related to metabolic-induced inflammation.Öğe Evaluation of Age Releated Anti-Müllerian Hormone Variations in Domestic Cat(2016) Yağcı, İlknur Pir; Polat, İbrahim Mert; Pekcan, MertAnti-Müllerian hormon (AMH) Transforming Growth Factor Beta ailesinin bir üyesidir ve dişilerde sadece ovaryumlardaki büyümekte olan granüloza hücrelerinden üretilir. Bu nedenle, ovaryumların varlığını ya da yokluğunu belirlemede AMH ölçümleri önemli rol oynamaktadır. Bu çalışmanın amacı evcil kedilerde ovariohisterektomi (OHE) öncesi ve sonrası bireysel AMH düzeylerinin değerlendirilmesi, OHE sonrası farklı günlerde alınan kan örneklerinde ortalama AMH değişimlerinin gösterilmesi, yaşa bağlı AMH düzeylerindeki değişimlerin belirlenmesi ve AMH düzeylerinin fonksiyonel ovaryum dokusunun belirlenmesinde bir tanı aracı olarak kullanılabilirliğinin araştırılmasıdır. Bu çalışmada toplam 30 evcil kedi kullanıldı. Hayvanlar yaşlarına göre iki gruba ayrıldı; <1 yaş (Grup 1, n=13), >1 yaş (Grup 2, n=17). Tüm kedilerin serum AMH konsantrasyonları 0 ve 3. günlerde sırasıyla 3.15±2.25 ng/mL ve 0.38±0.21 ng/mL olarak belirlendi. Operasyondan 10 gün sonra tüm kısırlaştırılmış kedilerin AMH düzeyleri ELISA kitinin ölçebildiği minimum değerden daha düşüktü (<0.08 ng/mL). Yaş sınıflandırmasına göre ise kedilerin yaşları ile AMH düzeyleri arasında 0. gün (rs=0.293, P<0.05) ve 3. günde(rs=0.410, P<0.01) korelasyon belirlendi. Sonuç olarak kedilerde fonksiyonel ovaryum dokusunun değerlendirilmesinde serum AMH konsantrasyonlarının ölçümü yeterli olacağı düşünüldü.Öğe Investigation of serum anti-Mullerian hormone levels at follicular phase and interestrus period in queens(Ankara Univ, 2024) Pir Yagci, Ilknur; Seven, Goekberk; Elifoglu, Taha Burak; Polat, Ibrahim Mert; Pekcan, MertThe objectives of this study were: i) to report overall mean AMH serum concentrations in queens, ii) to determine relationship between serum AMH concentration with estradiol level, iii) to investigate the relationship between serum AMH with follicular phase and interestrus phase of the estrous cycle, iv) to discuss the usability of AMH as a biomarker to diagnosis of ovarian remnant syndrome in breeding season in queens. In the study, 64 healthy queens referred to clinics for routine ovariohysterectomy were used. After anamnesis, vaginal cytology, macroscopic examination of the uterus and ovaries and estradiol measurement, queens were divided into two groups: follicular phase (n: 45) and interestrus (n: 19). Estradiol and AMH levels in serum samples were determined by ELISA. In the evaluation of serum AMH concentrations, a statistical difference was determined between the follicular phase and interestrus (P<0.05). A negative correlation was found between age and estradiol in both follicular phase (r = -0.171) and interestrus (r = -0.385) groups. A positive correlation was found between age and AMH in the interestrus (P<0.01, r = 0.696). Serum AMH levels in the interestrus were found to be significantly higher than follicular phase (P<0.05). It was thought that this increase in serum AMH levels during the interestrus period, which is the stage where oocytes were selected for ovulation, may be an indicator of the role of AMH in oocyte selection in queens, as in many other mammalian species.Öğe The Investigation of Serum Estrogen, Progesterone, AMH and IGF-1 Profiles in Lactating and Non Lactating Mares after Altrenogest Administrations(2020) Polat, İbrahim Mert; Korkmaz, Ömer; Elifoğlu, Taha Burak; Pir, İlknur Yağcı; Çolakoğlu, Hatice Esra; Pekcan, MertThe objective of this study is to follow up the follicular growth in ovaries until mating by ultrasonography to determine of pregnancy rates, serum estrogen, progesterone, AMH and IGF-1 concentrations in mares which synchronized with altrenogest and to compare the obtained data with non-lactating mares. The study was carried out during breeding season while ovaries were active; lactating mares were included in Group 1 (n = 8) and non-lactating mares in Group 2 (n = 8). In both groups, synchronization was performed by administration of altrenogest (Regumate®) and prostaglandin F 2 alpha (PGF2?). Dominant follicle diameters of non-lactating mares were found to be greater than those of lactating ones. During the study, estrogen, progesterone, AMH and IGF-1 levels were significantly lower in the lactating mares. The non- lactating mares were found to have a first pregnancy conception rate (75%), similar to that of lactating mares (62%) (x2=1.54, P=0.62). It was concluded that in the mares synchronized with altrenogest, the presence of lactation and the foal are significantly influences ovarian activities, ovarian steroid levels, AMH and IGF-1 levels. Therefore, in order to increase pregnancy rates in lactating mare, sexual cycle synchronization with altrenogest are considered to be a suitable method.
