Özet
Siklosporin A (CsA), klinik olarak bir bağışıklık baskılayıcı olarak yaygın şekilde kullanılmaktadır. Siklosporinin en önemli yan etkileri hipertansiyon, nefrotoksisite, hepatotoksisite, nörotoksisite ve hiperlipidemidir. N-asetilsistein (NAC) antioksidan etkileri kanıtlanmış bir maddedir. Bu çalışma siklosporin uygulanan ratlarda kan parametreleri üzerine NACnin etkilerini araştırmak amacıyla yapıldı. Bu amaçla 32 adet dişi rat her bir grupta 8 adet rat olacak şekilde, kontrol (standart şartlarda besleme, FTS) (1), NAC (100 mg/kg ?.P) (2), CsA (20 mg /kg S.C) (3), CsA (20 mg /kg S.C) + NAC (100 mg/kg ?.P) (4) olmak üzere 4 gruba ayrıldı. İki hafta süren bir denemenin sonunda ratların kalbinden alınan kanlardan elde edilen serumlarda aspartat aminotransferaz (AST) ve alanin aminotransferaz (ALT) aktiviteleri, üre, kreatinin, total protein, trigliserit total kolesterol, HDL-kolesterol, LDL-kolesterol düzeyleri ile tam kanda eritrosit (RBC), hematokrit (HCT), hemoglobin (HB), ortalama alyuvar hemoglobini (MCH), ortalama alyuvar hemoglobin konsantrasyonu (MCHC), ortalama alyuvar hacmi (MCV), lökosit (WBC), lenfosit (LYM), monosit (MON) ve nötrofil (NEU) değerleri belirlendi. Serum trigliserit düzeylerinin CsA grubunda diğer gruplara göre yüksek olduğu ve NAC verilmesinin CsA uygulamasıyla artan trigliserit düzeylerini azalttığı tespit edildi (P<0.001). LDL-kolesterol düzeylerinin CsA ve CsA+NAC gruplarında kontrol ve NAC gruplarına göre arttığı saptand (P<0.05). Hem CsA hem de NAC uygulamalarının diğer biyokimyasal parametrelerini gruplar arasında istatistiksel olarak anlamlı düzeyde etkilemediği görüldü (P>0.05). Lökosit (P<0.05) ve RBC (P<0.01) değerlerinin NAC grubunda diğer gruplara göre dü?ük olduğu belirlendi. Hemoglobin ve HCT değerlerinin NAC ve CsA gruplarında kontrol grubuna göre azaldığı tespit edildi (P<0.05). Sonuç olarak ratlara iki haftalık CsA uygulamasının trigliserit, LDL-kolesterol, HB ve HCT düzeyleri hariç diğer kan parametrelerini etkilemediği, 100 mg/kg NAC'nin ise CsA uygulaması ile artan trigliserit düzeylerini azalttığı, ancak diğer parametreleri anlamlı olarak etkilemediği tespit edildi. Ratlara uygulanan 20 mg/kg CsA?ya karşı NAC?nin farklı doz, verilme süreleri ve değişik formlarının kullanıldığı araştırmalarının yapılmasının faydalı olabileceği kanaatine varıldı.
Cyclosporine A (CsA) is widely used clinically as an immunosuppressant. The most important side effects of cyclosporine are hypertension, nephrotoxicity, hepatotoxicity, neurotoxicity and hyperlipidemia. N-acetylcysteine (NAC) is a substance with proven antioxidant effects. This study was carried out to investigate the effects of NAC on blood parameters in rats administered CsA. For this purpose, 40 male rats assigned to 4 groups, 8 animals in each group: control (standard feeding, FTS) (1), NAC (100 mg/kg ?.P) (2), CsA (20 mg /kg S.C) (3), CsA (20 mg /kg S.C) + NAC (100 mg/kg I.P) (4) (4). At the end of a two-week experiment, aspartate aminotransferase (AST) and alanin aminotrasferase (ALT) activities, total protein, urea, creatinine, triglyceride, total cholesterol, LDL-cholesterol, HDL-cholesterol levels were determined in the blood serum, whereas erythrocyte (RBC), hematocrit (HCT), hemoglobin (HB), mean red blood cell hemoglobin (MCH), mean red blood hemoglobin concentration (MCHC), mean red blood cell volume (MCV), leukocyte (WBC), lymphocyte (LYM), monocyte, neutrophil levels were determined in whole blood taken from the heart of rats. It was determined that serum triglyceride levels were higher in the CsA group than in the other groups and NAC administration reduced the triglyceride levels that increased with CsA administration (P<0.001). It was determined that LDL-cholesterol levels increased in the CsA and CsA+NAC groups compared to the control and NAC groups. It was observed that both CsA and NAC applications did not affect other biochemical parameters at a statistically significant level between the groups (P>0.05). Leukocyte (P<0.05) and RBC (P<0.01) values were determined to be lower in the NAC group compared to the other groups. It was determined that HB and HCT values decreased in the NAC and CsA groups compared to the control group (P<0.05).. As a result, it was determined that two-week CsA administration to rats did not affect other blood parameters except triglyceride, LDL-cholesterol, HB and HCT levels, and 100 mg/kg NAC reduced the triglyceride levels that increased with CsA administration, but did not significantly affect other parameters. It was concluded that it would be useful to conduct studies using different doses and different forms of NAC against 20 mg/kg CsA administered to rats.
