Özet
Bu çalışmaya Sağlık Bilimleri Üniversitesi, Kartal Şehir Hastanesi Patoloji Kliniğinde 2010 ve 2020 yılları arasında pleomorfik adenom, adenoid kistik karsinom ve mukoepidermoid karsinom tanısı almış 61 hasta dahil edildi. Hastaların demografik verileri ile bcl-2, kaspaz-3 ve GSTPi protein ekspresyonu immünohistokimyasal yöntemle incelendi ve karşılaştırıldı. Pleomorfik adenom tanılı 26 hastada GST izoenzimlerinden GSTA1 (alfa), GSTS1 (sigma), GSTT1 (teta), GSTM1 (mu), GSTO1 (omega), GSTK1 (kappa) protein ekspresyonları immünohistokimyasal yöntemle incelendi ve hastaların demografik verileri ile karşılaştırıldı. Bcl-2 boyanma yoğunluğu pleomorfik adenom tümör dokularına kıyasla adenoid kistik karsinom ve mukoepidermoid karsinom tümür dokuları arasında belirgin bir şekilde yüksek bulundu. Bcl-2 ekspresyonu ile hastaların demografik verileri kıyaslandığında ortalama tümör çapı ile hastaların bcl-2 ekspresyonu arasında anlamlı farklılık tespit edildi (p=0.045). Kaspaz-3 protein ekspresyonu ile bening ve malign tümörlü hastaların demografik verileri arasında anlamlı bir farklılık bulunamadı. GSTPi boyanma yoğunluğuna göre en yoğun boyanma mukoepidermoid karsinom tümör dokularında görüldü. GSTPi ekspresyonu ile hastaların demografik verileri arasında istatistiksel olarak anlamlı farklılık bulunamadı (p>0.05). Pleomorfik adenom tanısı almış 26 hastada, tümör dokuları ile tümöre komşu normal dokularda GSTA1 (alfa), GSTS1 (sigma), GSTT1 (teta), GSTM1 (mu), GSTO1 (omega), GSTK1 (kappa) protein ekspresyonları immünohistokimyasal yöntemle incelendiğinde; GSTA1 ve GSK1 protein ekspresyonu tümörlü dokulara kıyasla normal dokuda (p<0.05), GSTO1 ve GSTM1 protein ekspresyonu ise normal dokuya kıyasla tümörlü dokularda daha yüksek bulundu (p<0.05). Çalışmamız sonucunda benign tümörlere kıyasla malign tümörlerde daha güçlü bcl-2 boyanma yoğunluğu görüldü. In this study, were included 61 patients diagnosed with pleomorphic adenoma, adenoid cystic carcinoma, and mucoepidermoid carcinoma in the Pathology Clinic of the University of Health Sciences, Kartal City Hospital between 2010 and 2020. Demographic data of the patients and bcl-2, caspase-3, and GSTPi protein expression was analyzed and compared by immunohistochemical method. Protein expressions of GST isoenzymes GSTA1 (alpha), GSTS1 (sigma), GSTT1 (theta), GSTM1 (mu), GSTO1 (omega), GSTK1 (kappa) in 26 patients with a diagnosis of pleomorphic adenoma were analyzed by immunohistochemical method and compared with the demographic data of the patients. Bcl-2 staining intensity was significantly higher among adenoid cystic carcinoma and mucoepidermoid carcinoma tumor tissues compared to pleomorphic adenoma. When the bcl-2 expression was compared with the demographic data of the patients, a significant difference was found between the mean tumor diameter and the bcl-2 expression of the patients (p=0.045). There was no significant difference between caspase-3 protein expression and demographic data of patients with benign and malignant tumors. According to the GSTPi staining intensity, the most intense staining was seen in mucoepidermoid carcinoma tumor tissues. There was no statistically significant difference between GSTPi expression and demographic data of the patients (p>0.05). When the GSTA1 (alpha), GSTS1 (sigma), GSTT1 (theta), GSTM1 (mu), GSTO1 (omega), and GSTK1 (kappa) protein expressions in tumor tissues and normal tissues adjacent to the tumor in 26 patients diagnosed with pleomorphic adenoma were examined by immunohistochemical method; GSTA1 and GSK1 protein expression was found higher in normal tissues than tumor tissue (p<0.05). GSTO1 and GSTM1 protein expression were higher in tumor tissues than in normal tissue (p<0.05). As a result of our thesis study, bcl-2 staining intensity was observed to be higher in malignant tumors than benign tumors. When the mean tumor diameter and bcl-2 expression were compared, we found a significant difference between the mean tumor diameter and bcl-2 expression.
