Özet
Bu çalışmada, Kırıkkale Bölgesi'nde, perakende satış marketlerinden toplanan çeşitli pastörize sütlerde, izolasyon ve identifikasyonla Yersinia enterocolitica (Y. enterocolitica) varlığı ve serotiplerinin araştırılması planlanmıştır. Araştırmada izolasyon için alınan 100 adet pastörize süt numuneleri mikrobiyolojik analize alındı; 25 ml süt örneği, 225 ml Pepton Sorbitol Safra Suyu (PSBB) üzerine ilave edilerek, 30 sn homojenize edildi. İzolasyon için uygun besi yerine (Yersinia Selective Agar Base) ekim yapılarak, 10°C'de 10 gün inkübasyona bırakıldı. İnkübasyon sonucunda karakteristik özellik gösteren koloniler, hızlı identifikasyon sistemi (BD BBL Crystal E/NF) ile identifikasyona tabi tutuldu. İncelenen 100 adet pastörize sütte, Y. enterocolitica izole edilemedi; ancak 6 adet sütte maya, 1 adet sütte Shigella spp. tespit edildi. Daha yüksek sayıda numunenin çalışılmasının izolasyon oranını artırabileceği, önemli enfeksiyon kaynağı olan Y. enterocolitica'nın sütlerde incelenmesi; bir adet pastörize sütte izole edilen Shigella spp. tespit edilmesi halk sağlığı açısından önemlidir.
In this study, it was planned to investigate the presence and serotypes of Y. enterocolitica in various pasteurized milk collected from retail markets in Kırıkkale Region, with isolation and identification. 100 pasteurized milk samples taken for isolation in the study; 25 ml of milk sample was added to 225 ml of PSBB and homogenized for 30 seconds. Planting was done on suitable media (Yersinia Selective Agar Base) for isolation and incubated at 10°C for 10 days. Colonies showing characteristic features after incubation were identified by rapid identification system (BD BBL Crystal E/NF). Y. enterocolitica could not be isolated in 100 pasteurized milks examined, but yeast in 6 milks and Shigella spp. in 1 milk detected. It is thought that working with a higher number of samples may increase the isolation rate, and examining Y. enterocolitica, which is an important source of infection, in milk will be beneficial for public health. In addition, Shigella spp. detection is important for public health.
