Öğe Consequences of Social Isolation in Rats on Their Antioxidant Defense System and Erythrocyte Deformability(Aves, 2009) Aydogan, Sami; Ascioglu, Meral; Comu, Faruk MetinPurpose: In this study, our purpose was to investigate effect of oxidative stress composing in isolated rats on erythrocyte deformability and antioxidant system, considering importance of mechanical, and biochemical properties of erythrocytes in efficiency of blood circulation Material and Methods: Fourty Sprague-Dawley rats were divided into two groups according to their ages; where one of them consisted of 2 months old rats and the other 12 months old rats. Then, randomly selected ten rats in each group were isolated separately in a cage and hold 12/12 hour light-dark cycle lasting 21 days. Blood samples were collected at the end of the 21st day, and superoxide dismutasc (SOD), catalasc (CAT) activities and malondialdchydc (MDA) levels were measured spcctrophotometrically. Changes in plasma nitric oxide level were determined by Griess method depending on total nitrite -nitrate. In addition, relative filtration rate (RFR), relative filtration time (RFT) and relative resistance (Rrcl) of the erythrocytes were calculated as the indexes of erythrocyte deformability. Results: The SOD and CAT levels were found significantly lower in both of young and adult isolated rats when compared to the young and adult control groups. The nitric oxide levels were also found significantly lower in the young and adult isolated rats when compared to their control groups. The peroxidation of the lipids were also decreased by social isolation, in the erythrocyte membrane of both young and adult isolated rats compared to the controls. The indexes of erythrocyte deformability, RFR was found significantly higher, whereas the RFT was significantly lower in young and adult isolated rats compared to the young and adult controls. However, the Rrel of the erythrocytes were not altered dramatically. Conclusion: As a consequence, our data reveals that the social isolation causes a lack of plasma nitric oxide levels in the socially isolated rats and probably due to this decrease in one of the major oxidants for the erythrocyte membrane results in the decreased lipid peroxidation. Several cellular and tissue structural and functional alterations are observed in such injury. The aim of this study was to evaluate the effect of dexmedetomidine on lipid peroxidation and erythrocyte deformability during ischemia-reperfusion injury in rats. Methods: Twenty-four Wistar Albino rats were randomly separated into three groups as control (C), ischemia-reperfusion injury (I/R) and dexmedetomidine group (I/R-D). Ischemia was induced with portal clampage for 45 min and reperfusion period was 45 min after declampage. Group I/R-D received dexmedetomidine 100 mu g/kg i.p. 30 min before portal clampage. Serum malondialdehyde and superoxide dismutase activities to document lipid peroxidation and erythrocyte deformability index were investigated. Results: Serum superoxide dismutase and malondialdehyde activity levels were significantly higher and erythrocyte deformability index was decreased in hepatic ischemia-reperfusion group. However, these changes were observed to be prevented with dexmedetomidine treatment when given before portal clampage. Conclusion: These findings clearly indicate that erythrocyte deformability index is decreased in hepatic ischemia reperfusion injury and has a potential role to prevent these alterations. The protective effect of dexmedetomidine on hepatic I/R injury is also decreased lipid peroxidation. The aim of this study was to evaluate the effect of dexmedetomidine on ischemia-reperfusion injury (IRI) in rats with have hepatic IRI and diabetes mellitus. Methodology: Twenty-eight Wistar Albino rats were randomised into four groups as control (C), diabetic (DC), diabetic with hepatic ischemia-reperfusion injury (DIR), and diabetic but administered dexmedetomidine followed by hepatic IRI (DIRD) groups. Hepatic tissue samples were evaluated histopathologically by semiquantitative methods. Malondialdehyde (MDA), superoxide dismutase (SOD), glutathion s-transpherase (GST), and catalase (CAT) enzyme levels were investigated in liver and kidney tissues as oxidative state parameters. Results: In Group DIR; hepatocyte degeneration, sinusoidal dilatation, pycnotic nucleus, and necrotic cells were found to be more in rat hepatic tissue; while mononuclear cell infiltration was higher in the parenchyme. MDA levels were significantly lower; but SOD levels were significantly higher in Group DIRD with regard to Group DIR. In the IRI induced diabetic rats' hepatic and nephrotic tissues MDA levels, showing oxidative injury, were found to be lower. SOD levels, showing early antioxidant activity, were higher. Preoperative pregabalin has been used as a part of multimodal analgesia in postoperative pain treatment in recent years. Pregabalin has become one of the increasingly common agents in postoperative analgesia. In this study, we aimed to investigate the effect of pregabalin on erythrocyte deformability in rats undergoing IR. Methodology: 24 male Wistar albino rats weighing between 200-250 g were used in the study. Rats were randomly divided into 4 groups of 6 rats each (Control, Ischemia-Reperfusion (IR), IR-Pregabalin 50 mg (50 mg/kg), IR-Pregabalin 200 mg (200 mg/kg). Pregabalin was administered intraperitoneally 30 min before the procedure. An atraumatic microvascular clamp was placed across the infrarenal abdominal aorta in the IR groups. Following 120 min of ischemia, the clamp was removed and reperfusion was continued for 120 min. All rats were euthanized by intraperitoneal administration of ketamine (100 mg/kg) and taking blood from the abdominal aorta. Erythrocytes were seperated from heparinized whole blood samples. Deformability measurements were made in erythrocyte suspensions in phosphate buffered saline. A constant flow filtrometer system was used to measure erythrocyte deformability and relative resistance was calculated. Results: It was found that the formation of ischemia reperfusion increases the relative resistance according to the control group (p < 0.0001). It was determined that application of pregabalin 50 or 200 mg did not change erythrocyte deformability in ischemia reperfusion-induced rats (p = 0.632, p = 0.811). Conclusion: The administration of 50 or 200 mg of pregabalin has no negative effect on the erythrocyte deformability in ischemia reperfusion-induced rats. We think that pregabalin can be safely used for analgesia in the cases of IR. However, these findings should be supported by clinical and experimental studies carried out in more detailed and broader series.Öğe The effect of different types of honey on healing infected wounds(Ma Healthcare Ltd, 2018) Eroglu, Oguz; Deniz, Turgut; Kisa, Ucler; Comu, Faruk Metin; Kaygusuz, Sedat; Kocak, Orhan MuratObjective: To investigate the effects of treatments of 'mad honey', blossom honey and nitrofurazone on infected wound healing. Method: Male albino Wistar rats were randomly divided into four groups: 'mad honey' (MH), blossom honey (BH), nitrofurazone (N) and control (C). All rats were anaesthetised intraperitoneally. A circular skin incision was made to the back regions. Grafts containing slime-producing Staphylococcus epidermidis were placed on the incision area and then sutured to the skin. Infection in the wound area was confirmed after 48 hours. Wounds were dressed twice daily with the various treatment materials. Rats were randomly euthanised on days 7 or 14, and tissue samples taken. Tissue samples were assessed for hydroxyproline (HP), tensile strength (TS) and macroscopic measurement (area and intensity). Results: HP levels were higher in the treatment groups (MH, BH, N) at days 7 and 14 compared with the control group. 'Group x day' interaction was found in the HP levels (p=0.015). Increases in HP levels in the MH and N groups between days 7 and 14 were significantly higher than those in the other groups (p<0.05). Intensity was significantly lower in the control group and significantly higher in group MH compared with the other groups. Significant 'group x day' interaction was observed in intensity (p=0.006). TS was significantly lower on day 7 than on day 14 (p=0.022). No marked difference was observed between the groups, nor any 'group x day' interaction, in terms of TS. Conclusion: Honey administration successfully healed infected wounds. The aim of the present study was to examine the potential cardio protective effects of levosimendan in a diabetic rat model of myocardial I/R injury. Methods: A total of 18 streptozotocin-induced diabetic Wistar Albino rats (55 mg/kg) were randomly divided into three equal groups as follows: the diabetic I/R group (DIR) in which myocardial I/R was induced following left thoracotomy, by ligating the left anterior descending coronary artery for 60 min, followed by 2 h of reperfusion; the diabetic I/R levosimendan group (DIRL), which underwent I/R by the same method while taking levosimendan intraperitoneal 12 mu g kg(-1); and the diabetic control group (DC) which underwent sham operations without tightening of the coronary sutures. As a control group (C), six healthy age-matched Wistar Albino rats underwent sham operations similar to the DC group. Two hours after the operation, the rats were sacrificed and the myocardial tissue samples were examined by light microscopy for evidence of myonecrosis and inflammatory cell infiltration. Results: Myonecrosis findings were significantly different among groups (p = 0.008). Myonecrosis was more pronounced in the DIR group compared with the C, DC, and DIRL groups (p = 0.001, p = 0.007 and p = 0.037, respectively). Similarly, the degree of inflammatory cell infiltration showed significant difference among groups (p < 0.0001). Compared with C, DC, and DIRL groups, the inflammatory cell infiltration was significantly higher among the DIR group (p < 0.0001, p < 0.0001, and p = 0.020, respectively). Also, myocardial tissue edema was significantly different among groups (p = 0.006). The light microscopic myocardial tissue edema levels were significantly higher in the DIR group than the C, DC, and DIRL groups (p = 0.001, p = 0.037, and p = 0.014, respectively). Conclusion: Taken together, our data indicate that levosimendan may be helpful in reducing myocardial necrosis, myocardial inflammation, and myocardial tissue edema resulting from ischemia reperfusion injury.Öğe Effect of picroside II on erythrocyte deformability and lipid peroxidation in rats subjected to hind limb ischemia reperfusion injury(Dove Medical Press Ltd, 2016) Comu, Faruk Metin; Kilic, Yigit; Ozer, Abdullah; Kirisci, Mehmet; Dursun, Ali Dogan; Tatar, Tolga; Arslan, MustafaBackground: Ischemia reperfusion injury (I/R) in hind limb is a frequent and important clinical phenomenon. Many structural and functional damages are observed in cells and tissues in these kinds of injuries. In this study, we aimed to evaluate the effect of picroside II on lipid peroxidation and erythrocyte deformability during I/R in rats. Methods: Rats were randomly divided into four groups-each containing six animals (sham, I/R, sham + picroside II, and I/R + picroside II). The infrarenal section of the abdominal aorta was occluded with an atraumatic microvascular clamp in I/R groups. The clamp was removed after 120 minutes and reperfusion was provided for a further 120 minutes. Picroside II (10 mg.kg(-1)) was administered intraperitoneally to the animals in the appropriate groups (sham + picroside II, I/R + picroside II groups). All rats were euthanized by intraperitoneal administration of ketamine (100 mg.kg(-1)) and taking blood from the abdominal aorta. Erythrocytes were extracted from heparinized complete blood samples. Buffer (PT) and then erythrocytes (PE) were passed through the filtration system and the changes in pressure were measured to investigate the role of serum malondialdehyde and nitric oxide (NO) in lipid peroxidation and erythrocyte deformability index. Results: Deformability index was significantly increased in the I/R group compared to groups sham, sham + picroside-II, and I/R + picroside-II (P<0.0001, P<0.0001, and P=0.007). Malondialdehyde (MDA) and NO levels were evaluated. MDA level and NO activity were also higher in the I/R group than in the other groups. Picroside II treatment before hind limb I/R prevented these changes. Conclusion: These results support that deformability of erythrocytes is decreased in I/R injury and picroside II plays a critical role to prevent these alterations. Decreased erythrocyte deformability and increased aggregation may be detected after surgery performed under general anesthesia. It was shown that nonsteroidal anti-inflammatory drugs decrease erythrocyte deformability. Lornoxicam and/or intravenous (iv) ibuprofen are commonly preferred analgesic agents for postoperative pain management. In this study, we aimed to investigate the effects of lornoxicam (2 mg/kg, iv) and ibuprofen (30 mg/kg, iv) on erythrocyte deformability, as well as hepatic and renal blood flows, in male rats. Methods: Eighteen male Wistar albino rats were randomly divided into three groups as follows: iv lornoxicam-treated group (Group L), iv ibuprofen-treated group (Group I), and control group (Group C). Drug administration was carried out by the iv route in all groups except Group C. Hepatic and renal blood flows were studied by laser Doppler, and euthanasia was performed via intra-abdominal blood uptake. Erythrocyte deformability was measured using a constant-flow filtrometry system. Results: Lornoxicam and ibuprofen increased the relative resistance, which is an indicator of erythrocyte deformability, of rats (P=0.016). Comparison of the results from Group L and Group I revealed no statistically significant differences (P=0.694), although the erythrocyte deformability levels in Group L and Group I were statistically higher than the results observed in Group C (P=0.018 and P=0.008, respectively). Hepatic and renal blood flows were significantly lower than the same in Group C. Conclusion: We believe that lornoxicam and ibuprofen may lead to functional disorders related to renal and liver tissue perfusion secondary to both decreased blood flow and erythrocyte deformability. Methods: A total of 24 Wistar albino rats were divided into four groups: Control, sevoflurane, Alzheimer, and Alzheimer + sevoflurane. The experimental Alzheimer model was prepared by intracerebroventricular injection of streptozotocin at a dose of 3 mg/kg (10 mu l) to the Alzheimer and Alzheimer + sevoflurane groups. Sevoflurane (2.3%) was administered to the sevoflurane and Alzheimer + sevoflurane groups 4 weeks after the surgery for 3 days and lasting for 2 hours per day. Blood samples were then collected for deformability measurements. Results: The deformability index was significantly increased in Alzheimer-induced rats, but the results for the Alzheimer and Alzheimer + sevoflurane groups were similar. The erythrocyte deformability index was significantly increased in the Alzheimer and Alzheimer + sevoflurane groups (p=0.004 p=0.001 respectively). However, there was no difference in deformability in the sevoflurane group (p= 0.496) Conclusion: Erythrocyte deformability was increased in the Alzheimer-induced rats; however recurrent sevoflurane anesthesia did not affect erythrocyte deformability.Öğe The Effects of Simvastatin on Ischemia Reperfusion Injury in an Experimental Colon Anastomosis Model(Springer India, 2017) Akarsu, Mahmut; Saygun, Oral; Aydinuraz, Kuzey; Aydin, Oktay; Daphan, Cagatay Erden; Tanrikulu, Fatma Benli; Comu, Faruk MetinAnastomotic leakage is more frequently reported in colonic anastomoses. Ischemia reperfusion injury is one of the main reasons for anastomotic leakage. Simvastatin is known to prevent tissue damage induced by free oxygen radicals after ischemia reperfusion injury. The effect of simvastatin on colonic anastomosis impaired by ischemia reperfusion injury is investigated. Single layer, end-to-end colocolic anastomosis after 0.5-cm colon resection was performed in Wistar Albino rats. In Group 1 (control) (n= 10), colonic anastomosis without I-R was performed. In Group 2 (n = 10), the superior mesenteric artery was clamped for 10 min followed by 60 min of reperfusion after which resection anastomosis was performed. In Group 3 (n= 10), 10 mg/kg simvastatin was given by gavage for 7 days after I-R and resection anastomosis. In Group 4 (n = 10), the rats received 10 mg/kg simvastatin by gavage 7 days before and 7 days after ischemia reperfusion and surgery. All of the rats were sacrificed 8 days after surgery. Anastomotic bursting pressure and tissue hydroxyproline levels were measured. Postoperative administration of simvastatin restored the anastomotic bursting pressure and hydroxyproline levels to that of control group thus overcoming the effect of ischemia reperfusion injury. Simvastatin administered postoperatively in an experimental model of colonic resection anastomosis impaired by ischemia reperfusion injury increased anastomotic bursting pressures and tissue hydroxyproline levels. Further experimental and clinical studies will show whether administration of simvastatin will increase reliability of the anastomosis and decrease postoperative morbidity and mortality in colonic anastomosis after ischemia reperfusion injury.Öğe Effects of the general anaesthetic agent, propofol, on erythrocyte deformability(Comenius Univ, 2010) Arslan, Mustafa; Comu, Faruk Metin; Isik, Berrin; Unal, Yusuf; Cekmen, Nedim; Kurtipek, OmerObjectives: Propofol is an anesthetic agent frequently used for sedation and general anesthesia. The purpose of our study was to investigate the effect of propofol, a general anesthetic, on erythrocyte deformability in rats. Methodology: The study was performed on 20 male and 20 female rats, with 10 rats in each study group and 10 rats in each control group. The rats in the study group were administered propofol (150 mg.kg(-1)) intraperitoneally, and the rats in the control group were administered SF. Erythrocyte packs were prepared using heparinized total blood samples. Deformability measurements were done by erythrocyte suspensions in PBS buffer. A constant flow filtrometer system was used to measure erythrocyte deformability, and the relative resistance was calculated. Results: The use of propofol resulted in the increase in the relative resistance, which is an indicator for the erythrocyte deformability in both male and female rats (p=0.002, p=0.042, respectively). Conclusion: A negative change in the erythrocyte deformability may cause a functional deterioration in blood flow and tissue perfusion (Fig. 1, Ref. 23). This study aims to investigate the effects of Thymoquinone treatment on erythrocyte deform- ability in rats subjected to lower extremity ischemia-reperfusion injury. METHODS: The study was conducted on Wistar albino rats weighing 400-450 g. The rats were randomly divided into five groups: the control group (C), in which no treatment was applied; the group that received dimethyl sulfoxide (DMSO) as a solvent; the group subjected to 90 minutes of ischemia followed by 90 minutes of reperfusion in the main femoral artery of the lower extremity (IR); the Thymoquinone control group (TQ-C), in which the effects of Thymoquinone alone were examined; and the group that received intraperitoneal Thymoquinone one hour before the ischemia-reper fusion procedure (IR+TQ). At the end of the procedure, intracardiac blood was collected from the rats, and May-Grunwald and Giemsa (MGG) staining, endothelial nitric oxide synthase (eNOS), and erythrocyte deformability indexes were measured. RESULTS: The study results showed significant differences. Erythrocyte deformability was statistically significantly improved in the group that received Thymoquinone before ischemia-reper fusion compared to the group subjected to ischemia-reper fusion only. Morphological changes in erythrocytes were also statistically significantly better in the IR+TQ group than in the IR group. Immunohistochemical eNOS staining revealed that eNOS activity in the IR group was lower than in the IR+TQ group. Female (f) and male ( m) rats were divided into 3 groups; control (f (n = 7); m (n = 5)), sevoflurane treated group (f (n = 7); m ( n = 5)), desflurane treated group ( f ( n = 7); m (n = 7)). 2% of sevoflurane or 6% desflurane were applied to the rats with inhalation in an adjustable cage for one hour. The deformability indexes of the erythrocytes were measured by a laser diffractometer (Myrenne Rheodyne SSD). Sevoflurane anesthesia has improved the deformability of erythrocytes in male rats (p < 0.05) whereas there were not any significant changes in female rats. Desflurane has improved the deformability of erythrocytes in both gender significantly (p < 0.05). Volatil anesthetic agents sevofluran and desflurane has improved the mechanical properties of the erythrocytes in male rats compared to their controls. However, these changes were not significant with sevoflurane in females. The results in male rats may be due to the effects of testosterone on the flexibility of the erythrocytes leading them to tolerate to the environmental changes. These results reveal that the inhalation anesthetics like sevoflurane and desflurane are appropriate anesthetics which can improve the deformability of erythrocytes during surgery.
