Endemik Centaurea zaferii negaresh bitkisininantioksidan aktivitesi ve enzim inhibisyonetkilerinin belirlenmesi

Özet
Türkiye'de endemizm oranı %66 olan Centaurea L. cinsine ait birçok tür, halk arasında geleneksel olarak çok sayıda hastalığın tedavi edilmesinde kullanılmakta ve dolayısıyla bu türlerle ilgili biyoaktivite çalışmaları da giderek artmaktadır. Bu çalışmada kullanılan Centaurea zaferii Negaresh bitkisinden elde edilen farklı özütlerin toplam fenolik (TPC) ve flavonoid içeriklerinin (TFC) zengin, in vitro antioksidan ve diyabet ile ilişkili enzimler olan ?-amilaz ve ?-glukozidaz enzimlerini inhibe etme özelliklerinin güçlü olduğu gözlemlenmiştir. Maserasyon ile elde edilen metanol özütünde TPC (80,73 mg GAE/g) ve TFC (62,16 mg RE/g)' nin daha yüksek olduğu tespit edilmiştir. Diklorometan özütünün fosfomolibdenum (164,56 mg TE/g) antioksidan testinde ve ?-amilaz (490,67 mg ACE/g) inhibisyonunda, metanol ekstresinin ise DPPH (23,25 mg TE/g), ABTS (56,35 mg TE/g), CUPRAC (61,71 mg TE/g), FRAP (82,22 mg TE/g) antioksidan testlerinde ve ?-glukozidaz (1368,85 mg ACE/g) inhibisyonunda daha etkili olduğu belirlenmiştir. Sonuç olarak daha ileri fitokimyasal çalışmalar yapılarak C. zaferii Negaresh bitki özütlerinin içeriğinde bulunan etkin bileşiklerin tespit edilmesi ve bu özütlerin enzim inhibisyonu yanında farklı biyolojik etkinliklerinin belirlenip uluslararası bir dergide yayınlanması planlanmaktadır. Many species are belonging to the genus Centaurea, which the rate of endemism 61% in Turkey, used to treat many diseases traditionally among the people and thus the increasing bioactivity studies related to these species. Different extracts from Centaurea zaferii Negaresh plant used in this study were found to be rich in total phenolic (TPC) and flavonoid content (TFC), and the extracts were found to be strong in both in vitro antioxidant tests and inhibition of diabetes-associated enzymes (?amylase and ?-glucosidase). It was determined that TPC (80,73 mg GAE/g) and TFC (62,16 mg RE/g) was higher in methanol extract obtained by maceration. It was determined that dichloromethane extract was more effective in phosphomolybdenum (164,56 mg TE/g) antioxidant test and ?-amylase (490,67 mg ACE/g) inhibition, methanol extract in DPPH (23,25 mg TE/g), ABTS (56,35 mg TE/g), CUPRAC (61,71 mg TE/g), FRAP (82,22 mg TE/g) antioxidant tests, and ?-glucosidase (1368,85 mg ACE/g) inhibition. As a result, further phytochemical studies are planned to determine the active compounds in C. zaferii Negaresh plant extracts' content and determine the different biological activities of these extracts besides enzyme inhibition and to be published in an international journal.
